DNA of the tryptophan operon is transcribed in cytoplasmic extracts of E. coli. This transcription and its regulation by repression, occur in the absence of ribosomes. Repression requires the repressor protein product of the trp R gene and tryptophan; no involvement of the tryptophanyl-tRNA synthetase is found.
